Google has made the Panda Update twice (3.5 & 3.6) in the month of April. More over they have launched a new spam-fighting algorithm called Penguin. The Penguin update was targeting spam, you need to remove any spam you might have. In some cases, Google may have sent messages to you about spam activity in the past. Messages may even be waiting for you in Google Webmaster Central, if you’ve never verified your account.

Yes, the new update from Google is all about the spam content and spam links. therefore remove the spam, duplicate content from your site and remove all the spam links that might have for your website.

You may want to check the anchor text on your back links. If you have over optimized on your "main keyword" anchor text (meaning that 60% or more of your anchor text is your main keyword) the Penguin may penalize your ranking for that main keyword and maybe more.

    Penguin Update

Fortunately, none of my sites were affected, but I know people who took a hit. Since this Penguin Update seemed to target over optimized anchor text, that's where I looked first. I used Market Samurai in the SEO competition module for a particular keyword. Once I ran the SEO Competition analysis, there is a drop down for anchor text analysis for each URL. This lists the anchor text for that URL and how many back links are using that exact anchor text. The anchor text counts for each keyword should not be more than 60% of the total count. I wouldn't recommend removing any links, just begin to balance the numbers by adding anchor text links that may have low counts. Basically it shouldn't look like you have concentrated back links on any one keyword, keep it natural.

One of the many reasons could be the sites you are getting backlinks from got hit by the penguin updates and results in relative lowering (diluting) of your backlinks compared with your competitors.

Do you mean if I have a one-keyword-targeted website, I will lose PR?

No need to worry much this may happens but within a month will be back.. do SEO normally, do not do any black hat, dont make reconsideration request .. just wait a month after do anything if not back
