When you plug in a USB thumbdrive, select the last option - Take No Action.
In future when you plugin a USB drive, it will not pop up to open.
Or if you have already setup to open, you need go to My Computer - Right-click on the USB drive - Select Properties - AutoPlay - Choose Prompt me each time to choose an action.
Hope this helps!

My anti-virus actually will detect Virus on the USB drive if there is one. So, after that you can do the above as mentioned for future (in case your anti-virus does not detect immediately).

I suggest that you replace your anti-virus. Like Edhan said all anti-virus now detects viruses even without scanning. Like my anti-virus, even if I plug in my USB drive that has a virus and it automatically run it still detects the virus and prompt me for action. I suggest you get Kaspersky

Absent knowing what OS you are running, or if you are talking about a standalone machine, a server, or a network client, the best that I can offer at the moment, based on the assumption that you are running Windows, is to refer you to Microsoft Support: How to disable the Autorun functionality in Windows (http://support.microsoft.com/kb/967715).
