Default Does Javascript Produce Negative Affects

I am wondering if someone that is familiar with accessability could help me understand this better. I am wondering how the use of javascript affects page display and interpretation.

I have some javascript on certain segments of Website, but I believe they have passed a validation test. I noticed a couple posts concerning this issue and it peaked some concern.

the main issue is: does the site still work when javascript is not avaiable?
